A simple, green, and transition metal-free approach to the N-arylation of secondary amines with aryl halides in magnetized distilled water (MDW), as a solvent was introduced. This method offered the advantages of short reaction times, low costs, and an additive-free process, and it also enabled new planning strategies for the construction of aryl amine-containing pharmacophores. Also, the reaction rates were followed spectrophotometrically by monitoring the changes in the absorption spectrum of the reaction mixture in ordinary and magnetized distilled water. Furthermore, the reliable mol. dynamics (MD) simulations were used as predictive tools to interpret and describe the interactions between reactants in the ordinary and magnetized distilled water. The RDFs anal. of interactions revealed that morpholine and iodobenzene reactants were surrounded by more solvents in MDW than ordinary distilled water (ODW) 90°C. Consequently, the high reactivity observed for two reactants in MDW at 90°C can be explained by MD simulation, and there comes an established synergy between the theor. calculation and experiment

The structural design, preparation, spectral confirmation of thirteen new 4-ethoxyisophthalamide compounds I (R = 2-OEt, 3-Br, 4-I, etc.; n = 0, 1) were described and completes their preliminary screening of in vitro antiplatelet activity in response to the following inducers: ADP (ADP) and collagen. Among them, when ADP was used as an inducer, compounds having activities higher or close to picotamide and IC50 values revealed that one compound I (R = m-CF3, n = 0) (IC50 : 0.26μmol L-1) was more than three times more active than pos. control drug picotamide. Meanwhile, cytotoxic effects in vitro of compound I (R = 4-I, n = 0) and compound I (R = m-CF3, n = 0) on L929 cells were analyzed. The structure-activity relationship showed that when electron-withdrawing groups nitro or trifluoromethyl are introduced at the meta- or para- position of the two side chain benzene rings, it is beneficial on the whole to the improvement of antiplatelet activity.

The phthalazine core in LY2940680 was replaced with anthranilamide, which retained the inhibitory activity towards the hedgehog (Hh) signaling pathway as evidenced by a dual luciferase reporter gene assay. Compound I (R = Ph; R1 = 2-CF3-4-F; R2 = CH) displayed the best inhibitory activity against the Hh signaling pathway with IC50 value of 34.09 nM, and exhibited better proliferation inhibitory activity towards the Daoy cell line (IC50 = 0.48μM) than LY2940680 (IC50 = 0.79μM).

In this study, we designed and synthesized a series of I [R = H, 2-OH, 3-OH, 4-OH,..etc]. as potent Hsp90 inhibitors by simplifying and modifying the aconitine scaffold. Among these compounds, I [R = 3-Br] exhibited an excellent antiproliferative activity against LoVo cells with an IC50 value of 0.02μM and a significant Hsp90α inhibitory activity with an IC50 value of 0.71 nM. Mol. docking studies provided a rational binding model of I [R = 3-Br] in complex with Hsp90α. The following cell cycle and apoptosis assays revealed that compound I [R = 3-Br] could arrest cell cycle at G1/S phase and induce cell apoptosis via up-regulation of bax and cleaved-caspase 3 protein expressions while inhibiting the expressions of bcl-2. Moreover, I [R = 3-Br] could inhibit cell migration in LoVo and SW620 cell lines. Consistent with in vitro results, I [R = 3-Br] significantly repressed tumor growth in the SW620 xenograft mouse model.

All the synthesized compounds I were tested for their in vitro antiproliferative activity against two breast cancer cell lines (MDA-MB-231 and MCF7), antibacterial and antifungal activities against tested strains. Among the tested compounds, compound I [R = F] observed as highest potent candidate against MDA-MB-231with IC50 of 5.44 μg/mL and compound I [R = Cl] was observed as most potent candidate against Gram-neg. bacterial strains.

Parasitic roundworms (nematodes) are significant pathogens of humans and animals and cause substantive socioeconomic losses due to the diseases that they cause. The control of nematodes in livestock animals relies heavily on the use of anthelmintic drugs. However, their extensive use has led to a widespread problem of drug resistance in these worms. Thus, the discovery and development of novel chem. entities for the treatment of parasitic worms of humans and animals is needed. Herein, authors describe the medicinal chem. optimization efforts of a phenotypic hit against Haemonchus contortus based on a pyrrolidine-oxadiazole scaffold I (R = H, 4-Cl, 3-OMe, etc.; R1 = 2-Me, 3-OH, 4-CN, etc.). This led to the identification of compounds with potent inhibitory activities (IC50 = 0.78-22.4μM) on the motility and development of parasitic stages of H. contortus, and which were found to be highly selective in a mammalian cell counter-screen. These compounds could be used as suitable chem. tools for drug target identification or as lead compounds for further optimization.

In base-free conditions, it has been proved that the yield could achieve to 99.9% for the synthesis of benzimidazoles with liberating water as the sole byproduct, and the catalyst remains stable and efficient even after five successive cycle tests. Addnl., experiments and characterizations confirm the good oxidation activity of the catalyst benefits from the numerous oxygen vacancies provided by the high concentration of low-valent cobalt (Co2+) and the doping of carbon improves the intermol. transport of electrons. Furthermore, this strategy could potentially be applied in the industrial production of benzimidazoles.
