Reaction between acyl chloride and substituted aromatic amines for the synthesis of new diamide compounds I [R = Ph, 2-FC6H4, 4-BrC6H4, etc.] was investigated. Compounds I were evaluated for 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) and ferrous ions (Fe2+) chelating activities in terms of antioxidant properties. The IC50 value was calculated for the compounds which showed activation in preliminary study and pos. controlled BHT, BHA and trolox. Compounds I were evaluated for antibacterial activity against Escherichia coli ATCC 25922, Staphylococcus aureus ATCC 25923, Enterococcus fecalis ATCC 29212, Pseudomonas aeruginosa ATCC 27853 and Enterobacter aerogenes ATCC 13048 and it was determined that compounds I [R = Ph, 2-F3CC6H4] showed activity close to pos. control against S. aureus and E. aerogenes bacteria, resp.

The X-ray data showed that compounds I [R = Me, Cl] contain two mols. and compound I [R = F] contain one mol. in asym. unit. Comparison of conformation of two distinct mols., in the asym. unit of compounds I [R = Me, Cl] were studied with aid of reported literature. All the synthesized compounds I were tested for their in vitro antiproliferative activity against two breast cancer cell lines (MDA-MB-231 and MCF7), antibacterial and antifungal activities against tested strains. Among the tested compounds, compound I [R = F] observed as highest potent candidate against MDA-MB-231with IC50 of 5.44 μg/mL and compound I [R = Cl] was observed as most potent candidate against Gram-neg. bacterial strains.

In this study, we designed and synthesized a series of I [R = H, 2-OH, 3-OH, 4-OH,..etc]. as potent Hsp90 inhibitors by simplifying and modifying the aconitine scaffold. Among these compounds, I [R = 3-Br] exhibited an excellent antiproliferative activity against LoVo cells with an IC50 value of 0.02μM and a significant Hsp90α inhibitory activity with an IC50 value of 0.71 nM. Mol. docking studies provided a rational binding model of I [R = 3-Br] in complex with Hsp90α. The following cell cycle and apoptosis assays revealed that compound I [R = 3-Br] could arrest cell cycle at G1/S phase and induce cell apoptosis via up-regulation of bax and cleaved-caspase 3 protein expressions while inhibiting the expressions of bcl-2. Moreover, I [R = 3-Br] could inhibit cell migration in LoVo and SW620 cell lines. Consistent with in vitro results, I [R = 3-Br] significantly repressed tumor growth in the SW620 xenograft mouse model.

A broad range of substrates including heterocycles, chalcones and sterically hindered coupling partners were well tolerated in the developed protocol. Addnl., the metal complex formed in the catalytic cycle was characterized by 1H-NMR spectroscopy and mass spectrometry.

An economically efficient and environmentally benign approach for the direct oxidative transformation of aldehydes to nitriles was developed using com. available non-toxic copper acetate as an inexpensive catalyst and ammonium acetate as the source of nitrogen in the presence of aerial oxygen as an eco-friendly oxidant under ligand-free conditions. The reactions were associated with high yield and various sensitive moieties like allyloxy, benzyloxy, t-butyldimethylsilyloxy, hetero-aryl, formyl, keto, chloro, bromo, methylenedioxy and cyano were well tolerated in the aforesaid method. The kinetic studies showed first order dependency on the aldehyde substrate in the reaction rate. The reaction was faster with the electron deficient aldehydes as confirmed by Hammett anal. Moreover, the present oxidative method was effective on larger scales showing potential for industrial application.
