The discovery, from nature, of a large and diverse set of nitrilases is reported. The utility of this nitrilase library for identifying enzymes that catalyze efficient production of valuable hydroxy carboxylic acid derivatives is demonstrated. Unprecedented enantioselectivity and substrate scope are highlighted for 3 newly discovered and distinct nitrilases. For example, a wide array of (R)-mandelic acid derivatives and analogs were produced with high rates, yields, and enantiomeric excesses (95-99% ee). We also have found nitrilases that provide direct access to (S)-phenyllactic acid and other aryllactic acid derivatives, again with high yields and enantioselectivities. Finally, different nitrilases have been discovered that catalyze enantiotopic hydrolysis of 3-hydroxyglutaronitrile to afford either enantiomer of 4-cyano-3-hydroxybutyric acid with high enantiomeric excesses (>95% ee). The 1st enzymes are reported that effect this transformation to furnish the (R)-4-cyano-3-hydroxybutyric acid, which is a precursor to the blockbuster drug Lipitor.

A study on the hierarchical self-assembly behavior of the obtained metallacage revealed that it displayed a solvent-induced emission switch accompanied by enhancement of the emission intensity as a result of the change in intermol. Pt···Pt and π-π interactions. More interestingly, the metallacage has been found to spontaneously self-assemble into a transparent metallogel at room temperature without a heating-cooling process.

This method takes advantage of the nitro group as a traceless activating and directing group (TADG) that is released in the aromatization step to produce 4-substituted 3-perfluoroalkyl pyrazoles with complete regioselectivity [e.g, trans-β-nitrostyrene + CF3CHN2 → pyrazole I (89%) in presence of Ag2O and Na3PO4]. The potential of this method is further demonstrated by the synthesis of penthiopyrad. Safety: 2,2,2-trifluorodiazoethane is potentially explosive.

It concentration-dependently attenuated the calcium channel current and extracellular Ca2+ influx, without affecting the intracellular Ca2+ mobilization, in vascular smooth muscle cells from rats. 16C, possessing the N-4-tert-Bu benzyl piperidine structure, as a novel calcium antagonist, may be effective as a calcium channel blocker in cardiovascular disease.

ERK1 and ERK5 are proposed to have pivotal roles in several types of cancer. Under some circumstance, ERK5 may provide a common bypass route, which rescues proliferation upon abrogation of ERK1 signaling. Thus, we accurately classified the tumor types from The Cancer Genome Atlas (TCGA) based on the expression levels of ERK1 and ERK5. We proposed a novel therapeutic strategy to overcome the above-mentioned compensatory mechanism in specific tumor types by co-targeting both ERK1 and ERK5. On the basis of the idea of overcoming ERK5 compensation mechanism, 22ac (ADTL-EI1712) as the first selective dual-target inhibitor of ERK1 and ERK5 was discovered to have potent antitumor effects in vitro and in vivo. Interestingly, this compound was found to induce regulated cell death accompanied by autophagy in MKN-74 cells. Taken together, our results warrant the potential of this dual-target inhibitor as a new candidate drug that conquers compensatory mechanism in certain tumor types.
